چکیده
T lymphocytes accumulate in inflamed tissues of patients with chronic inflammatory diseases (CIDs) and express pro-inflammatory cytokines upon re-stimulation vitro. Further, a significant genetic linkage to MHC genes suggests that play an important role the pathogenesis CIDs including juvenile idiopathic arthritis (JIA). However, functions established disease remain elusive. Here we dissect transcriptional clonal heterogeneity synovial JIA by single-cell RNA sequencing combined cell receptor profiling on same cells. We identify clonally expanded subpopulations expressing reflecting recent activation antigen situ. A PD-1+TOX+EOMES+ population CD4+ expressed immune regulatory chemoattractant for myeloid PD-1+TOX+BHLHE40+ CD4+, mirror CD8+ driving inflammation, supporting B lymphocyte This analysis points out multiple types have be targeted therapeutic regeneration tolerance arthritis.

PD1 signal transduction pathways in T cells
The use of immune checkpoint inhibitors for the treatment of cancer is revolutionizing oncology. Amongst these therapeutic agents, antibodies that block PD-L1/PD1 interactions between cancer cells and T cells are demonstrating high efficacies and low toxicities. Despite all the recent advances, very little is yet known on the molecular intracellular signaling pathways regulated by either PD-L1 ...
متن کاملA Threshold Level of Intratumor CD8+ T-cell PD1 Expression Dictates Therapeutic Response to Anti-PD1.
Despite successes, thus far, a significant proportion of the patients treated with anti-PD1 antibodies have failed to respond. We use mouse tumor models of anti-PD1 sensitivity and resistance and flow cytometry to assess tumor-infiltrating immune cells immediately after therapy. We demonstrate that the expression levels of T-cell PD1 (PD1(lo)), myeloid, and T-cell PDL1 (PDL1(hi)) in the tumor m...

Background Multicentric carpotarsal osteolysis (MCTO), a skeletal dysplasia presents in early childhood mimicking juvenile idiopathic arthritis (JIA). Recognition of this syndrome is essential to avoid unnecessary treatment with immunosuppressive agents because of different course and treatment. Case Report A 3-year-old boy presented with swelling and restriction of right wrist joint and left ...
